Content Manager is a service within the Cognos 8 architecture.  Content store is a database used to hold the Cognos 8 content.

As for the final question - it is far too vague to provide any meaningful answer.  It's rather like asking "How do I cook?" :)

Cognos Content Manager:
Cognos content manager is the Cognos 8 Service which manages the storage of Application Data.
This includes security, Configuration data, Models, Matrix, Reports   Specification and Report Outputs.
Needed to publish models, retrieve or store reports specification and their outputs.
Needed for Manage Scheduling information and Manage Cognos Namespace.

Content Store:
The Cognos 8 service that uses a content store is named as content     manager.
The content store is a relational database that contains data which Cognos 8 needs to operate.
It contains report specifications, publish models and the packages, connection information of data sources, information about external namespace or information about scheduling and bursting report.
A Cognos 8 content store can be an MS SQL Server, Oracle, Sybase,DB2 or any other database.
For supporting Multilingual the oracle database parameter NLS character set  should be AL32UTF 8.
Much of the information in the content store, such as report results and    XML report specifications, is stored as binary large object (BLOB) fields.
